　　幕末时期，四维动荡。新撰组虽然网罗四方高手，但已有名无实。适值此时，来自盛冈南部藩的吉村贯一郎（中井贵一 饰）加入新撰组，其为人油滑悭吝，然非凡的剑术与缜密精细的头脑令土方岁三（野村祐人 饰）、斋藤一（佐藤浩市 饰）、冲田总司（堺雅人 饰）等名侍也不由得暗自钦佩。随着倒幕运动初露端倪，新撰组统归幕府直接领导，其血腥无情的作风更被世人称作“壬生之狼”。值此关隘，伊东甲子太郎（斋藤步 饰）等人不愿沦为幕府走狗，反出新撰组。最强大的武士部队分裂，与之相伴的必然是一场混杂着阴谋和杀戮的腥风血雨……
